Triple Reporter Virus

A triple reporter virus is an engineered virus that produces three distinct measurable signals, enabling researchers to monitor multiple aspects of viral infection in the same experimental system. Its genome contains separate reporter gene cassettes that are expressed during infection, generating signals such as fluorescence, bioluminescence, or enzymatic activity according to the construct design. In immunology and infection research, these viruses can help track infection sites, compare viral replication with host responses, and evaluate antiviral treatments. Multiparameter reporting provides richer spatial and temporal data than a single reporter, supporting more precise analysis of virus-host interactions and disease mechanisms.

We describe the usage of a fluorescent reporter vaccinia virus that enables real-time measurement of viral infectivity and gene expression through the stage-specific expression of spectrally distinct reporter fluorophores. We detail a plate-based method for accurately identifying the stage at which virus replication is affected in response to small molecule inhibition.
