Aspartate Aminotransferase

Aspartate aminotransferase (AST) is an enzyme that transfers amino groups between amino acids and keto acids, supporting cellular amino acid metabolism and serving as a clinically useful marker of tissue injury. In a pyridoxal phosphate-dependent transamination reaction, AST converts aspartate and alpha-ketoglutarate into oxaloacetate and glutamate. AST is abundant in the liver, heart, skeletal muscle, kidneys, and red blood cells, so elevated blood levels can indicate damage in these tissues. In medicine, AST measurement is commonly interpreted with alanine aminotransferase and other laboratory findings to assess liver injury, monitor disease, and distinguish potential sources of cellular damage.

Over the recent years, live cell-based assays have been used successfully to detect antibodies against surface and conformational antigens. Here, we describe a method using high-throughput flow cytometry enabling the analysis of large cohorts of patients. Detection of novel antibodies will improve diagnosis and treatment of immune-mediated disorders.

Here, we describe the use of a genetically directed alkaline phosphatase (AP) labeling method to investigate the anatomical features and phenotypes of mouse retinas under NMDA-induced excitotoxicity. In conjunction with several retinal ganglion cell (RGC) subtype-specific reporter lines, we uncover novel phenotypes in dying RGCs throughout the degenerative process.

We ectopically expressed NR1 subunit of NMDA receptor tagged with green fluorescent protein in human embryonic cells (HEK293) as antigen to detect autoantibodies against NMDA receptor in the blood of patients suspected with autoimmune encephalitis. This simple method may be suitable for screening purposes in clinical settings.

Relying on meticulous anatomical landmarks without intraoperative ultrasound, this approach offers a precise and parenchyma-sparing option for dorsally located right-liver tumors, particularly in settings with limited advanced navigation resources.
