Bone Mesenchymal Stem Cells

Bone mesenchymal stem cells are multipotent stromal cells found in bone marrow that support skeletal tissue maintenance and repair, making them important in regenerative medicine. Their behavior depends on signals from the surrounding marrow niche, which can direct self-renewal and differentiation into osteoblasts, chondrocytes, or adipocytes through changes in gene expression and cell signaling. Researchers study these cells to understand bone development, fracture healing, and skeletal disease, while investigating their potential for tissue engineering, cell-based therapies, and drug testing. Their accessibility and differentiation capacity make them valuable tools for connecting basic stem cell biology with clinical applications.

An Enzymatic Method to Rescue Mesenchymal Stem Cells from Clotted Bone Marrow Samples

Mesenchymal stem cells are usually obtained from bone marrow and require expansion culture. When samples clot before processing, a protocol using the (enzymatic) thrombolytic drug urokinase can be applied to degrade the clot. Thus, cells are released and available for expansion culture. This protocol provides a rapid and inexpensive alternative to resampling.

This protocol describes the radiolabeling of equine mesenchymal stem cells and their implantation into tendon injuries in the horse in order to determine cell survival and tissue distribution using gamma scintigraphy.

A protocol establishing mesenchymal stem cells (MSCs) isolated from the synovial fluid and bone marrow of minipigs and non-invasively collected using syringe aspiration is presented. Cellular phenotyping was performed using flow cytometry after cell isolation and in vitro cultivation of bone marrow and synovial fluids derived from MSCs.

Age-related diseases are associated with multiple defects in components of the cytoplasm. Here, we present a protocol to prepare plasma membrane vesicles from bone marrow mesenchymal stem cells. This technique could potentially be used as a means of cytoplasm replacement therapy to ameliorate or even reverse age-associated phenotypes.
