Contrast Medium Perfusion

Contrast medium perfusion is an imaging approach that uses an injected contrast agent to visualize and measure blood flow through organs and tissues, helping reveal vascular function that structural images alone may miss. As the contrast bolus passes through the circulation, changes in X-ray attenuation, magnetic resonance signal, or ultrasound echoes are recorded over time; these data generate time–concentration curves and quantitative measures such as tissue blood volume and flow. In medicine, the technique supports assessment of cerebral ischemia, myocardial perfusion, tumors, and other conditions involving altered vascular supply, improving diagnosis, treatment planning, and response monitoring.

Here, we present a protocol to inject ultrasound microbubble contrast agents into living, isolated late-gestation stage murine embryos. This method enables the study of perfusion parameters and of vascular molecular markers within the embryo using contrast-enhanced high-frequency ultrasound imaging.
