Dopamine Depletion

Dopamine depletion is a reduction in the availability or activity of dopamine, a neurotransmitter involved in movement, motivation, reward, and several hormonal functions. It can result from impaired dopamine synthesis, reduced release, accelerated breakdown, or loss of dopamine-producing neurons, particularly within the nigrostriatal pathway that supports coordinated movement. In medicine, dopamine depletion is strongly associated with Parkinson’s disease, where degeneration of these neurons contributes to tremor, rigidity, and bradykinesia. Measuring or experimentally inducing depletion helps researchers study neurological disorders, while treatments such as levodopa aim to restore dopamine signaling and improve motor function.

Measuring Dopamine Uptake in Mouse Brain Synaptosomes Using Radiolabeled Dopamine

2025

This video demonstrates the assessment of dopamine uptake in synaptosomes harvested from mouse brains. It outlines steps for preparing synaptosomes for dopamine uptake via dopamine transporters, treating them with varying concentrations of radiolabeled dopamine, isolating the synaptosomes, and measuring radioactivity to quantify dopamine uptake.

Creating Dynamic Images of Short-lived Dopamine Fluctuations with lp-ntPET: Dopamine Movies of Cigarette Smoking

2013

We present a novel PET imaging approach for capturing dopamine fluctuations induced by cigarette smoking. Subjects smoke in the PET scanner. Dynamic PET images are modeled voxel-by-voxel in time by lp-ntPET, which includes a time-varying dopamine term. The results are 'movies' of dopamine fluctuations in the striatum during smoking.

Isolation and Culture of Primary Embryonic Mouse Midbrain Dopamine Neurons

2025

The video demonstrates the process of isolating and culturing dopamine neurons from the midbrain of mouse embryos. The isolated ventral midbrain floors are dissociated using enzymes. The resulting dopamine neurons are harvested and transferred into micro-islands inside multi-well plates. The cells are incubated with a dopamine neuron medium to maintain the cells at a high density inside the micro-islands.
