Glomeruli Counting

Glomeruli counting is the quantitative assessment of glomeruli, the kidney’s capillary-rich filtration units, in tissue sections or whole organs. In medicine and renal research, investigators use microscopy, image analysis, and systematic sampling to identify and enumerate glomerular profiles, often reporting glomerular density or total number. Accurate counts depend on clear tissue preparation, representative sampling, and consistent criteria for distinguishing glomeruli from surrounding structures. This measurement supports evaluation of nephron endowment, kidney development, and pathological changes such as glomerular loss or remodeling, helping relate structural alterations to renal function and disease progression.

Glomeruli Isolation: A Technique to Obtain Intact Glomeruli from Murine Kidney

2023

This video describes the method to isolate intact glomeruli from mouse kidneys. The encapsulated and decapsulated glomeruli thus obtained can further be used for culturing parietal epithelial cell outgrowths from the murine kidney.

Single-channel Analysis and Calcium Imaging in the Podocytes of the Freshly Isolated Glomeruli

Changes in the intracellular calcium levels in the podocytes are one of the most important means to control the filtration function of glomeruli. Here we explain a high-throughput approach that allows detection of real-time calcium handling and single ion channels activity in the podocytes of the freshly isolated glomeruli.

Using a Hemacytometer to Count Cells

2023

Many biomedical experiments require manipulation of a known quantity of cells, in order to achieve accurate, reproducible, and statistically-relevant data. Therefore, learning how to count cells is a particularly essential technique for any successful biomedical scientist. The most common way to count cells is by using a hemacytometer - an instrument that bears two laser-etched grids, which aid in the enumeration of an aliquot cells under a simple light microscope. This data can then be used to...

Knowledge of the exact number of viable cells is required for many tissue culture manipulations. This protocol describes how to differentiate between live and dead cells and quantify cells using a hemacytometer. Although it describes counting human neural stem/precursor cells (hNSPCs), it can be used for other cell types.
