Gst Fusion

GST fusion is a recombinant protein strategy in which glutathione S-transferase is genetically linked to a protein of interest, enabling its production and purification for biomedical research. During expression, the GST tag can improve protein solubility, and the resulting fusion protein binds specifically to immobilized glutathione for affinity chromatography; cleavage methods can then remove the tag when required. In medicine, GST fusion supports the preparation of purified antigens, enzymes, receptors, and other research proteins used in biochemical assays, structural studies, antibody development, and investigations of disease-related molecular interactions.

Affinity Precipitation of Active Rho-GEFs Using a GST-tagged Mutant Rho Protein (GST-RhoA(G17A)) from Epithelial Cell Lysates

The method presented here describes an assay to follow activation of RhoA specific GDP/GTP Exchange Factors (GEFs) in cultured cells by making use of a mutant RhoA GST fusion protein that has high affinity for activated GEFs. GEFs are precipitated from cell lysates, detected by Western blotting and quantified by densitometry.

In the present protocol, we demonstrate a highly efficient and cost-effective small-scale protein purification method, which allows purification of recombinant proteins by uniquely combining a cleavable GST-tag and a small His-tag.

Here, we present a protocol for live imaging of mouse secondary palate fusion using confocal microscopy. This protocol can be used in combination with a variety of fluorescent reporter mouse lines, and with pathway inhibitors for mechanistic insight. This protocol can be adapted for live imaging in other developmental systems.
