Rat Liver Model

A rat liver model is an experimental system that uses rat liver tissue or whole animals to investigate hepatic function, disease, and responses to treatment. Because the liver regulates metabolism, detoxification, protein synthesis, and bile production, researchers can examine how genetic, chemical, surgical, or infectious challenges alter hepatocytes and tissue organization. These models support studies of liver injury, regeneration, fibrosis, cancer, drug metabolism, and toxicity, while enabling measurements of biochemical, cellular, and physiological outcomes. In medicine, rat liver models help evaluate disease mechanisms and therapeutic strategies before clinical research, although differences between rat and human biology require careful interpretation.

We describe a method to produce an animal model of liver fibrosis in the rat, and assess the degree of fibrosis by histological examination of the liver. The model can be used to study the development of liver disease as well as to test the efficacy of potential anti-fibrotic agents.

Source: Hiller, T., et al. Study of Viral Vectors in a Three-dimensional Liver Model Repopulated with the Human Hepatocellular Carcinoma Cell Line HepG2. J. Vis. Exp., (2016)This video demonstrates adeno-associated virus-mediated gene delivery in a humanized rat liver scaffold. The vector delivers a transgene encoding a fluorescent reporter and gene-silencing RNA into liver cells via receptor-mediated endocytosis. This model supports efficient transduction in a perfused 3D liver tissue system.

Inducing rapid liver hypertrophy using Associating Liver Partition and Portal vein ligation for a Staged hepatectomy (ALPPS) has been proposed for resection of borderline resectable liver tumors. This model may elucidate mechanisms involved in rapid hypertrophy and allows testing of drugs that promote or block the acceleration of regeneration.
