Intestinal Injury

Intestinal injury is damage to the structure or function of the small or large intestine, potentially disrupting digestion, absorption, and the protective barrier between the gut and surrounding tissues. It can result from inflammation, infection, ischemia, mechanical trauma, or exposure to toxic substances, with epithelial disruption allowing fluid loss and altered interactions between intestinal microbes and immune cells. In medicine, assessing intestinal injury supports diagnosis and management of conditions such as inflammatory bowel disease, intestinal obstruction, and treatment-related toxicity. Research on epithelial repair, barrier integrity, and tissue regeneration may improve biomarkers, therapies, and recovery strategies.

Here we describe the detailed procedure of intestinal ischemia-reperfusion in mice which results in reproducible injury without mortality to encourage the standardization of this technique across the field. This model of intestinal ischemia-reperfusion injury can be utilized to study the cellular and molecular mechanisms of injury and regeneration.

Important Endpoints and Proliferative Markers to Assess Small Intestinal Injury and Adaptation using a Mouse Model of Chemotherapy-Induced Mucositis

Here, we present a protocol to establish important endpoints and proliferative markers of small intestinal injury and compensatory hyperproliferation using a model of chemotherapy-induced mucositis. We demonstrate the detection of proliferating cells using a cell cycle specific marker and using small intestinal weight, crypt depth, and villus height as endpoints.

We describe how to generate a widely used surgical model of intestinal ischemia-reperfusion injury (IRI) in rodents. The procedure involves occlusion of the superior mesenteric artery followed by the restoration of blood flow. This model is useful for studies investigating occlusive causes of intestinal IRI in both veterinary and human medicine.
