Lps Antibody Complex

An LPS antibody complex is an immune complex formed when an antibody binds lipopolysaccharide (LPS), an endotoxin on the outer membrane of Gram-negative bacteria. Antibody recognition targets specific LPS structures, including lipid A or polysaccharide regions, which can limit endotoxin interaction with immune receptors such as Toll-like receptor 4 and promote clearance through Fc-receptor-mediated uptake or complement activation. In medicine, LPS antibody complexes support research on sepsis, endotoxemia, and host-pathogen interactions, while related antibodies contribute to diagnostic assays, immunological studies, and investigations of strategies to reduce excessive inflammation caused by bacterial infection.

Described here is a technique in which lipopolysaccharide is injected into the lactating mouse mammary gland via the nipple to simulate mastitis, a condition commonly caused by bacterial infection. Lipopolysaccharide injection results in increased nuclear factor kappa B (NF-κB) signaling, visualized through bioluminescent imaging of an NF-κB luciferase reporter mouse.

A Serum Bactericidal Assay for the Complement-Mediated Bactericidal Activity of Antibodies

This video demonstrates a technique to evaluate the complement-mediated bactericidal activity of antibodies present in serum. This approach combines isolated serum with pathogenic bacteria and exogenous complement proteins. Upon incubation, the bacteria are grown on an agar plate, and the produced colonies are quantified to measure the serum bactericidal titer.

In this study, we describe an improved protocol for a multiplexed high-throughput antibody microarray with lectin detection method that can be used in glycosylation profiling of specific proteins. This protocol features new reliable reagents and significantly reduces the time, cost, and lab equipment requirements as compared to the previous procedure.
