Lung Histopathology

Lung histopathology is the microscopic examination of lung tissue to identify structural changes associated with disease, injury, infection, or cancer. After tissue fixation, sectioning, and staining, typically with hematoxylin and eosin, researchers and pathologists assess alveoli, airways, blood vessels, interstitium, and inflammatory or neoplastic cells, linking cellular patterns to underlying mechanisms. In medicine, this approach supports diagnosis, disease classification, treatment planning, and evaluation of therapeutic responses across conditions such as pneumonia, fibrosis, emphysema, and lung tumors. Quantitative image analysis and specialized stains or immunohistochemistry can further clarify tissue composition, disease progression, and clinically relevant biomarkers.

Non-Invasive Endotracheal Administration of Lipopolysaccharide to Induce Acute Lung Injury in Rodents

2025

In this study, a non-invasive endotracheal instillation technique is demonstrated to develop ALI in rats using a basic laryngoscope to ensure precise LPS delivery to the lungs. The LPS is instilled via an endotracheal tube using a laryngoscope. The magnitude of lung injury is evaluated at multiple time points.

Picrosirius Red Staining for Semiquantitative Histopathologic Evaluation of Collagen Deposition in Murine Models of Chronic Lung Allograft Rejection

Picrosirius Red staining is a semiquantitative method for objectively assessing collagen deposition in murine pulmonary fibrotic remodeling in a spatially resolved manner. Apart from evaluating total collagen deposition, Picrosirius Red staining allows for differentiating collagen fibers of different thicknesses.

This paper describes a striaghforward and efficient method of intubating mice for pulmonary function measurements or pulmonary instillation, that allows the mice to recover and be studied at later times. The procedure involves an inexpensive fiberoptic light source that directly illuminates the trachea.

2013

A method to image ex vivo pulmonary resection specimens with optical frequency domain imaging (OFDI) and obtain precise correlation to histology is described, which is essential to developing specific OFDI interpretation criteria for pulmonary pathology. This method is applicable to other tissue types and imaging techniques to obtain precise imaging to histology correlation for accurate image interpretation and assessment. Imaging criteria established with this technique would then be...
