Microbubble Contrast Agent

A microbubble contrast agent is an injectable suspension of gas-filled microspheres used to enhance the visibility of blood flow and tissue perfusion during medical imaging, especially contrast-enhanced ultrasound. When exposed to ultrasound waves, the microbubbles oscillate because their compressible gas cores respond differently from surrounding tissue, producing strong echoes that improve image contrast. Their circulation within the bloodstream can help clinicians assess cardiac function, vascularity, organ perfusion, and selected lesions in real time. Because the bubbles remain largely within the vascular space and are cleared from the body, they provide a valuable alternative or complement to other imaging contrast media.

This protocol describes the preparation and characterization of a dendrimeric magnetic resonance imaging (MRI) contrast agent that carries cyclen-based macrocyclic chelates coordinating paramagnetic gadolinium ions. In a series of MRI experiments in vitro, this agent produced an amplified MRI signal when compared to the commercially available monomeric analogue.
