Murine Inducible Alleles

Murine inducible alleles are engineered mouse gene variants whose activity can be switched on or off at a chosen time, allowing researchers to study gene function in living animals while limiting developmental or systemic effects. They typically use conditional DNA elements, such as loxP sites recognized by Cre recombinase, or drug-responsive regulatory systems activated by compounds including tamoxifen or doxycycline; the trigger controls recombination or gene transcription in selected tissues. In medicine, these models help link genes to disease onset, progression, and treatment response, supporting studies of cancer, immunity, metabolism, and regenerative biology.

Telomere and telomerase play essential roles in ageing and tumorigenesis. The goal of this protocol is to show how to generate two murine inducible telomerase knock-in alleles and how to utilize them in the studies of tissue degeneration/regeneration and cancer.

Murine Model of Epicutaneously-Induced Immunomodulation

2025

The study demonstrates the role of the skin in modulating immune responses in mice through epicutaneous (EC) immunization. EC immunization with a protein antigen can suppress immune responses in contact hypersensitivity or reverse skin-induced suppression when combined with pathogen-associated molecular patterns. EC immunization shows promise as an effective, needle-free approach to immunotherapy.

Multiple Allele Traits

For the same gene multiple alleles can interact to influence phenotypes like the shape and protein composition of an individual cells.By studying allele interactions on the molecular and cellular levels researchers can understand the resulting phenotypes and complications of human conditions like sickle cell trait, improving treatment. The ABO blood group system is a common example of multiple alleles in humans. This system includes three alleles called IA, IB, and i alleles, which combine in...

Lethal Alleles

Agouti: A Lethal Allele Lucien Cuénot discovered lethal alleles in 1905 while studying the inheritance of coat color in mice. The agouti gene is responsible for the color of the coat in mice. This gene codes for an agouti-signaling protein, which is responsible for melanin distribution in mammals. The wild-type allele gives rise to gray-brown coat color in mice, while the mutant allele gives rise to yellow coat color. In addition to coat color, the agouti gene is associated with the yellow...

A Method to Induce Ocular Surface Inflammation in a Murine Model

2025

This video demonstrates a method to induce ocular surface inflammation and meibomian gland dysfunction in a murine model. Stimulation of the mouse's immune system using an immunogen and later restimulation with the same immunogen promotes ocular surface inflammation, which causes plugging of meibomian gland secretion through neutrophil-produced aggNETs and leads to dry eye disease.
