Msc Seeding

MSC seeding is the controlled placement of mesenchymal stem cells onto culture surfaces, biomaterials, or tissue-engineering scaffolds to establish a viable and evenly distributed cell population. The process depends on cell–surface adhesion, seeding density, scaffold architecture, and culture conditions, which influence attachment, survival, proliferation, and subsequent differentiation. In medicine, MSC seeding supports the development of engineered tissues and cell-based models for regenerative research, including investigations of bone, cartilage, and other damaged tissues. Optimizing seeding efficiency can improve cellular integration, reproducibility, and the therapeutic potential of biomaterial-based approaches.

MSC Seeded Scaffold Preparation: A Technique to Prepare Scaffold Seeded with Mesenchymal Stem Cells for Surgical Implantation into Murine Model

This video describes the technique of seeding mesenchymal stem cells (MSCs) onto a biodegradable poly(lactic acid) (PLA) scaffold. The prepared scaffolds can be implanted into the surgical resection cavity of the brain from where they may migrate towards tumor foci. Further, these cells can be engineered to express therapeutic proteins to stimulate apoptosis in cancer cells.

MitoCeption: Transferring Isolated Human MSC Mitochondria to Glioblastoma Stem Cells

Here, a protocol (MitoCeption) is presented to transfer mitochondria, isolated from human mesenchymal stem cells (MSC), to glioblastoma stem cells (GSC), with the goal of studying their biological effects on GSC metabolism and functions. A similar protocol can be adapted to transfer mitochondria between other cell types.

Herein, we communicate protocols to efficiently generate and administer therapeutic spherical aggregates or 'spheroids' of MSCs primed under xeno-free conditions for experimental and clinical applications.
