Murine Aorta Isolation

Murine aorta isolation is a microsurgical dissection technique used to remove the aorta from a mouse for ex vivo vascular research and analysis. The procedure relies on careful separation of the vessel from surrounding connective tissue, blood, and branching vessels while preserving the aortic wall and its cellular architecture; the isolated tissue can then be mounted, cultured, fixed, or processed for molecular and histological studies. Flow Cytometry Analysis of Immune Cells Within Murine Aortas

This paper presents a flow cytometry-based method to investigate the immune composition of aortas. The paper also illustrates an additional technique that allows examining surrounding adventitia and vessel wall separately. This method opens possibilities to perform phenotypical analyses of aortic leukocytes and apply several immunological assays for atherosclerosis studies.

In this video, we demonstrate a procedure for the surgical removal of the aorta from miniature pigs. Endothelial cells harvested from these isolated aortae can be further cultured in in vitro conditions to study coagulation and immune responses.
