Oleic Acid Induction

Oleic acid induction is an experimental method used to create acute lung injury, particularly in preclinical medical research. The procedure introduces oleic acid into the circulation, where the fatty acid injures pulmonary vascular endothelium, increases capillary permeability, and promotes alveolar edema, inflammation, and impaired gas exchange. This model can reproduce key features of acute respiratory distress and related pulmonary dysfunction, allowing researchers to investigate injury mechanisms, monitor physiological changes, and evaluate potential drugs or supportive treatments. Its controlled, reproducible nature makes oleic acid induction valuable for studying vascular leak and lung injury, although findings require careful interpretation when translated to human disease.

The present protocol describes a lung injury model in mice using oleic acid to mimic acute respiratory distress syndrome (ARDS). This model increases the inflammatory mediators on edema and decreases lung compliance. Oleic acid is used in the salt form (oleate) since this physiological form avoids the risk of embolism.
