Tissue Sampling

Tissue sampling is the collection of cells or tissue from the body for microscopic, molecular, or other laboratory analysis, helping clinicians identify disease and guide treatment. Samples may be obtained through methods such as needle aspiration, core biopsy, endoscopic biopsy, or surgical excision, with the selected approach depending on the tissue’s location, size, and clinical question. In the laboratory, specimens are preserved, prepared, stained, and examined for structural or molecular abnormalities. Tissue sampling supports diagnosis, cancer classification, infection assessment, and evaluation of treatment response, while careful collection and handling help preserve specimen quality and improve the reliability of medical decisions.

Quantitation of DNA double-strand breaks on the basis of γH2AX foci has become an invaluable tool, particularly in radiation biology, for the evaluation of tissue radiosensitivity and effects of radiation modifying compounds. Here we demonstrate the use of an immunofluorescence assay for quantitation of γH2AX foci in tissue samples.

A Hydrophobic Tissue Clearing Method for a Rat Brain Tissue Sample

Source: Kirchner, K.N., et al., A Hydrophobic Tissue Clearing Method for Rat Brain Tissue. J. Vis. Exp. (2020).This video demonstrates the method of hydrophobic tissue clearing of a rat brain tissue sample. The sample is sequentially dehydrated in methanol, treated with a dichloromethane and methanol mixture to dissolve lipids and pigments, and transferred to dibenzyl ether for complete tissue clearing, enabling detailed imaging without thin sectioning.

Dissociating and Culturing Neurons from Hippocampal Tissue Samples

This video demonstrates the dissociation and culture of hippocampal neurons from hippocampal tissue samples using enzymatic and mechanical dissociations. The treatment with Ara-C inhibits the growth of non-neuronal cells, facilitating the selective growth of hippocampal neurons.

Solid Phase Microextraction Sampling: A Probe-based Technique for Sample Extraction from Graft Tissues
