3d Spheroid Model

A 3D spheroid model is a laboratory-grown, three-dimensional aggregate of cells that more closely reproduces aspects of tissue organization than conventional two-dimensional cultures. In neuroscience, neural cells are assembled or maintained under conditions that limit attachment to a flat surface, allowing cell-cell interactions and extracellular-matrix signaling to promote compact structures with internal gradients of oxygen, nutrients, and metabolites. These models support studies of neural development, differentiation, connectivity, neurodegeneration, and responses to candidate therapies. By capturing selected features of brain tissue while remaining experimentally accessible, 3D spheroids can complement animal studies and improve the physiological relevance of in vitro research.

To study ovarian tumor progression in a physiologically relevant model, multicellular spheroids were cultured in a microdevice under simulated fluid flow. This dynamic 3D model emulates the intraperitoneal environment with the cellular and mechanical components where ovarian cancer metastasis occurs.

This protocol describes the technique for performing extracellular flux assay on 3D spheroids generated from the co-culture of pancreatic cancer cells (Patu8902) and activated pancreatic fibroblasts (PS1) to measure the two major energy-producing pathways, glycolysis and mitochondrial respiration.

Spheroid Generation from Cell Lines: A Three-Dimensional (3D) Cell Culture Method

Spheroids are a type of 3D cell culture model used to more accurately represent a cell’s environment compared to 2D cell culture monolayer models. In the example protocol, we will see scientists generate spheroids from a colorectal cancer cell line and apply live-cell imaging to track the spheroids’ growth.
