Awake Mouse Recording

Awake mouse recording is a neuroscience technique for measuring brain activity in mice that remain conscious during observation, allowing neural signals to be related to natural behavior. Researchers use implanted electrodes, fiber photometry, or optical imaging to detect electrical or calcium-dependent activity while the animal is head-fixed or freely moving, often alongside behavioral tracking. This approach avoids the widespread effects of anesthesia on neuronal circuits and sensory processing. Awake recordings help investigators examine neural coding, perception, learning, movement, and disease-related changes, providing a closer link between cellular activity and behavior in health and neurological research.

Electrophysiological characterization of neuronal responses is important for understanding brain function and for guiding the placement of dyes for pathway tracing. However, many studies are performed in anesthetized animals. To understand brain function without anesthetics, we developed a method to record neuronal response properties and inject dyes in awake mouse.

(2015)This video demonstrates the method of juxtacellular recording and staining in a restrained, awake rat. The procedure involves recording signals from neurons and injecting dye to precisely localize the recording site. This technique allows accurate mapping of neural activity to the brain's anatomical structure.

The design and assembly of microdrives for in vivo electrophysiological recordings of brain signals from the mouse is described. By attaching microelectrode bundles to sturdy driveable carriers, these techniques allow for long-term and stable neural recordings. The lightweight design allows for unrestricted behavioral performance by the animal following drive implantation.
