Hek Cell Aggregation

HEK cell aggregation is the clustering of human embryonic kidney (HEK) cells into multicellular groups, a culture behavior relevant to organizing cells for in vitro experiments. It occurs when cell-cell adhesion and physical contact overcome forces that keep cells dispersed, with aggregation influenced by culture conditions such as cell density, medium composition, and agitation. In neuroscience, researchers use HEK cells as tractable heterologous systems to study neuronal receptors, ion channels, and signaling proteins; controlling aggregation can affect cell-cell interactions, experimental uniformity, and the interpretation of functional assays.

Rapid Isolation of the Mitoribosome from HEK Cells

Mitochondria have specialized ribosomes that diverged from their bacterial and cytoplasmic counterparts. Here we show how mitoribosomes can be obtained from their native compartment in HEK cells. The method involves isolation of mitochondria from suspension cells and consequent purification of mitoribosomes.

4D Imaging of Protein Aggregation in Live Cells

Cellular viability depends on timely and efficient management of protein misfolding. Here we describe a method for visualizing the different potential fates of a misfolded protein: refolding, degradation, or sequestration in inclusions. We demonstrate the use of a folding sensor, Ubc9ts, for monitoring proteostasis and aggregation quality control in live cells using 4D microscopy.

Tracking Hypoxic Signaling within Encapsulated Cell Aggregates

A method for photo-encapsulation of cells in a crosslinked PEG hydrogel is described. Hypoxic signaling within encapsulated murine insulinoma (MIN6) aggregates is tracked using a fluorescent marker system. This system allows serial examination of cells within a hydrogel scaffold and correlation of hypoxic signaling with changes in cell phenotype.
