Localizer Scan Procedure

A localizer scan procedure is a neuroimaging method used to identify brain regions that respond selectively to a particular stimulus, task, or sensory feature. In neuroscience, it commonly uses task-based functional magnetic resonance imaging to detect blood-oxygen-level-dependent signals as neural activity alters local blood flow and oxygenation; researchers then compare responses across experimental conditions. The resulting functional map can define regions of interest for later analyses, reduce anatomical ambiguity, and support individualized investigation of perception, language, memory, and other cognitive processes. Localizer scans therefore connect experimental tasks with the brain circuits that support them.

This paper introduces an approach of combining laser scanning photostimulation with whole cell recordings in transgenic mice expressing GFP in limited inhibitory neuron populations. The technique allows for extensive mapping and quantitative analysis of local synaptic circuits of specific inhibitory cortical neurons.

We describe a method to localize fluorescently tagged proteins in electron micrographs. Fluorescence is first localized using photo-activated localization microscopy on ultrathin sections. These images are then aligned to electron micrographs of the same section.
