Ex-vivo Tissue Imaging

Ex vivo tissue imaging is the visualization of biological structures and molecular signals in tissue removed from an organism, preserving spatial relationships for detailed analysis. In neuroscience, researchers prepare brain or nervous tissue by maintaining it viable or fixing and labeling it, then use microscopy to capture fluorescence, morphology, and optical sections across regions or cellular layers. This approach reveals neuronal organization, synaptic architecture, vascular patterns, and responses to experimental conditions that may be difficult to resolve in living animals. Ex vivo imaging supports studies of neural development, connectivity, disease mechanisms, and the effects of drugs or genetic manipulation.

The assembly and use of a multimodal microendoscope is described which can co-register superficial tissue image data with tissue physiological parameters including hemoglobin concentration, melanin concentration, and oxygen saturation. This technique can be useful for evaluating tissue structure and perfusion, and can be optimized for individual needs of the investigator.

Mesoscopic fluorescence tomography operates beyond the penetration limits of tissue-sectioning fluorescence microscopy. The technique is based on multi-projection illumination and a photon transport description. We demonstrate in-vivo whole-body 3D visualization of the morphogenesis of GFP-expressing wing imaginal discs in Drosophila melanogaster.

Ex Vivo Infection of Live Tissue with Oncolytic Viruses

Oncolytic viruses are promising for cancer therapeutics. The ability to ascertain the infectability of live tissue specimens obtained from patients prior to treatment is a unique advantage of this therapeutic approach. This protocol describes how to process tissues for ex vivo infection with oncolytic virus and subsequent viral quantification.
