Mouse Embryo Culture

Mouse embryo culture is an ex vivo technique that maintains developing mouse embryos under controlled laboratory conditions, allowing researchers to examine embryogenesis without relying solely on in vivo experiments. Embryos are placed in nutrient-defined culture media and maintained at controlled temperature and gas conditions so that cell division, tissue organization, and neural development can continue outside the uterus. In neuroscience, this approach supports direct analysis of neural tube formation, brain patterning, neuronal differentiation, and responses to genetic or environmental perturbations. It provides a valuable model for studying developmental mechanisms, identifying causes of neurodevelopmental abnormalities, and evaluating candidate interventions.

This video demonstrates the ex vivo culture and imaging of oculomotor slices from transgenic mouse embryos. The embryos are isolated and embedded in agarose. Using a vibratome, slices containing the oculomotor nuclei and eyes are obtained. The slices are maintained in culture and imaged using a fluorescence microscope.

Whole embryo culture technique allows us to culture mouse and rat embryos ex vivo condition during limited periods corresponding to midgestation stages. In this video protocol, we demonstrate our standard procedures of rat whole embryo culture after E12.5 using the rotator-type bottle culture system.
