Neuron-specific Promoter

A neuron-specific promoter is a regulatory DNA sequence that drives gene expression preferentially in neurons, making it a key tool for targeting genetic activity within the nervous system. It works when transcription factors and other regulatory proteins recognize promoter elements and recruit RNA polymerase, initiating transcription in suitable neuronal cell types while limiting expression in non-neuronal cells. In neuroscience, these promoters can control transgenes encoding fluorescent reporters, optogenetic actuators, or therapeutic proteins in cultured neurons, animal models, and gene-delivery systems. Their selectivity helps researchers map neural circuits, examine neuronal function, and reduce unintended effects, although promoter activity can vary with cell type, developmental stage, and experimental context.

Promoter expression analyses are crucial to improving the understanding of gene regulation and the spatiotemporal expression of target genes. Herein we present a protocol to identify, isolate, and clone a plant promoter. Further, we describe the characterization of the nodule-specific promoter in the common bean hairy roots.

Using a Macrophage Conditioned Medium to Promote Neuron Extensions

This video demonstrates an assay for promoting the growth of neuron projections known as neurite outgrowth using the macrophage-conditioned medium. Co-culture neurons and macrophages with cAMP and harvest the macrophage-derived conditioned medium. Growing the neurons with the conditioned medium promotes neurite outgrowths. These outgrowths are fluorescently labeled and visualized using a fluorescence microscope.

This procedure yields telencephalic neurons by going through checkpoints which are similar to those observed during human development. The cells are allowed to spontaneously differentiate, are exposed to factors which push them towards the neural lineage, are isolated, and are plated onto coverslips to allow for terminal differentiation and maturation.
