Synaptic Connection Mapping

Synaptic connection mapping is the systematic identification and characterization of communication links between neurons, revealing how neural circuits are organized. Depending on the question, researchers trace axons and dendrites with anatomical labeling, visualize synaptic contacts using microscopy, or test functional connectivity through electrophysiological recording and targeted stimulation. These complementary approaches distinguish structural pathways from synapses that actively transmit signals, helping relate circuit architecture to neuronal activity and behavior. In neuroscience, synaptic connection mapping supports studies of sensory processing, learning and memory, disease mechanisms, and potential circuit-based therapies.

Studying Synaptic Vesicle Pools using Photoconversion of Styryl Dyes

FM dyes have been of invaluable help in the understanding of synaptic dynamics. FMs are normally followed under the fluorescent microscope during different stimulation conditions. However, photoconversion of FM dyes combined with electron microscopy allows the visualization of distinct synaptic vesicle pools, among other ultrastructure components, in synaptic boutons.

Mapping Inhibitory Neuronal Circuits by Laser Scanning Photostimulation

This paper introduces an approach of combining laser scanning photostimulation with whole cell recordings in transgenic mice expressing GFP in limited inhibitory neuron populations. The technique allows for extensive mapping and quantitative analysis of local synaptic circuits of specific inhibitory cortical neurons.

This protocol describes the quantification of volumetric cerebral hemodynamic variations in the mouse brain using functional ultrasound (fUS).
