Thermode Application Protocol

Thermode Application Protocol is a standardized procedure for delivering controlled thermal stimuli to the skin during neuroscience experiments, enabling precise assessment of sensory and pain responses. A temperature-controlled thermode makes contact with a defined skin region and applies programmed warming or cooling, activating peripheral thermoreceptors and nociceptors while researchers record detection thresholds, ratings, or reflexive responses. The protocol supports investigations of somatosensory processing, pain pathways, and neural responses to temperature. By controlling stimulus intensity, rate, duration, and location, it improves reproducibility across participants and helps evaluate sensory function, experimental interventions, and changes associated with neurological conditions.

We describe a modified DIG in situ hybridization protocol, which is fast and applicable on a wide range of plant species including Norway spruce. With just a few adjustments, including altered RNase treatment and proteinase K concentration, the protocol may be used in studies of different tissues and species.

The development of new ultrasound (US) probes based on Capacitive Micromachined Ultrasonic Transducer (CMUT) technology requires an early realistic assessment of imaging capabilities. We describe a repeatable experimental protocol for US image acquisition and comparison with magnetic resonance images, using an ex vivo bovine brain as an imaging target.
