Tissue Equilibration Protocol

A tissue equilibration protocol is a standardized procedure that allows excised or isolated tissue to reach stable physiological and experimental conditions before pharmacological measurements begin. During equilibration, the tissue is maintained in an appropriate medium under controlled temperature, gas, pH, and buffer conditions so that diffusion, metabolism, and recovery from dissection approach a consistent baseline. This preparation improves the reliability and reproducibility of responses to drugs, agonists, or antagonists in organ bath and ex vivo studies. Consistent equilibration is therefore essential for comparing tissue contractility, receptor activity, dose-response relationships, and drug effects across experiments.

A simple liquid nitrogen Dewar/cryostat apparatus comprised of a small fused silica optical Dewar, a thermocouple, and a charge-coupled device (CCD) spectrograph are described. The experiments for which this Dewar/cryostat is designed require fast sample loading, freezing, and alignment, accurate and stable sample temperatures, and small size/portability.
