Southern Blot

サザンブロッティングは、標識DNAプローブが標的DNAとハイブリダイズして、ゲノム内の特定の配列を検出する技術です。

まず、ゲノムDNAを制限酵素で消化し、得られたDNA断片をアガロースゲルにロードし、ゲル電気泳動を用いて分離します。
二本鎖DNAを変性または2本鎖DNAまたはssDNAに分離するために、ゲルを水酸化ナトリウム溶液に浸します。
次に、ゲルを塩化ナトリウムとトリス緩衝液を含むDNA中和溶液のスポンジに置き、pHを7.0にリセットします。
次に、ナイロンメンブレンをゲルの上に置き、ペーパータオルの束で重さを量ります。ssDNAは毛細管現象によって膜上に移動し、中和溶液中の高塩濃度はDNAの結合を助けます。

Southern blotting is a DNA detection method used after agarose gel electrophoresis separates DNA fragments by size. A DNA ladder with fragments of known length is run beside the sample so the fragment size can be estimated. The blot adds another step when scientists need to confirm the sequence identity of the DNA.

First, the target DNA is cut into smaller pieces with a restriction endonuclease and run on an agarose gel. The DNA is then denatured, which means the double strands are separated into single strands. These fragments are transferred from the gel onto a nylon or nitrocellulose membrane so they can be reached by a probe.

A probe is a small single-stranded DNA fragment that is complementary to the target sequence and carries a reporter tag. On the membrane, the DNA is immobilized by UV irradiation for nylon or by heat for nitrocellulose. Nylon membranes are used more often because baking nitrocellulose at 80 °C can be hazardous.

The membrane is exposed to the labeled probe, which basepairs with any matching DNA sequence to form a hybrid molecule. Extra probe that does not bind is washed away. The hybridization pattern is then detected, often by autoradiography, to show where the target DNA is present.

Southern blots help identify DNA and measure its size and abundance. They are also used to study deletions, insertions, and rearrangements in genes. In some cases, they can detect a point mutation if it changes a restriction site. A related method, Northern blotting, is used for RNA and is often used to detect gene expression by measuring the mRNA transcript.

Investigation of techniques utilized could come from field trips to witness the use of these techniques in practice, lab exercises, virtual labs, simulation activities, interviews with professionals, etc. Molecular techniques could include: PCR, electrophoresis, restriction enzyme digestion of DNA, DNA sequencing, plasmid-based transformation, transfection, etc. These techniques are used in fields of medicine, agriculture, biomedical engineering, forensic science, etc.

Students should begin to appreciate the differences in ethical values that exist, and recognize that discussion of these values is imperative as knowledge and technology continue to advance, even when resolutions of differences can be rare.
