Natural Products Biosynthesis

Natural products biosynthesis is the enzyme-mediated production of specialized organic molecules by living organisms, including plants, fungi, bacteria, and marine organisms. It begins with primary metabolic building blocks that enzymes transform through reactions such as condensation, oxidation, reduction, methylation, glycosylation, and cyclization, often across coordinated gene-encoded pathways. These pathways generate chemically diverse compounds involved in ecological interactions and organismal adaptation, while also providing sources and design principles for pharmaceuticals, agrochemicals, fragrances, and other bioactive products. In biochemistry, studying their enzymes, intermediates, and pathway regulation supports natural product discovery, metabolic engineering, and sustainable production.

The heterologous biosynthesis of erythromycin A through E. coli includes the following experimental steps: 1) genetic transfer; 2) heterologous reconstitution; and 3) product analysis. Each step will be explained in the context of the motivation, potential, and challenges in producing therapeutic natural products using E. coli as a surrogate host.

This article provides a detailed protocol for the preparation and evaluation of monoclonal antibodies against natural products for use in various immunoassays. This procedure includes immunization, cell fusion, indirect competitive ELISA for positive clone screening, and monoclonal hybridoma preparation. The specifications for antibody characterization using MALDI-TOF-MS and ELISA analyses are also provided.

Here, we present a protocol for direct, early stage guanidinylation that enables rapid total synthesis of aminoguanidine-containing small organic molecules. An advanced synthetic intermediate used in the synthesis of a blood coagulation factor XIa inhibitor was prepared using this protocol.
