Payload Delivery

Payload delivery is the process of transporting a defined cargo, such as a drug, nucleic acid, protein, or imaging agent, to a desired location while preserving its activity and controlling its release. In bioengineering, delivery systems may encapsulate or chemically link the payload to a carrier that improves stability, directs tissue or cellular uptake, and releases the cargo in response to conditions such as pH, enzymes, or external stimuli. Effective payload delivery supports drug development, gene therapy, diagnostics, and regenerative medicine by increasing local efficacy and reducing unintended exposure, while advances in carrier design aim to improve precision, safety, and therapeutic outcomes.

The blood-brain barrier is a significant hurdle in the delivery of therapies for glioblastoma, a disease for which there is no cure. Here, we report an in vivo image-guided iron oxide therapeutic nano platform that can bypass this physiological barrier by virtue of size and accumulate in the tumor.

Drug targeting to central nervous system tumors is a major challenge. Here we describe a protocol to produce an in vitro mimic of the blood-brain tumor-barrier using murine and/or human cells and discuss their relevance for the predictability of central nervous system tumor targeting in vivo.

Insonation of microbubbles is a promising strategy for tumor ablation at reduced time-averaged acoustic powers, as well as for the targeted delivery of therapeutics. The purpose of the present study is to develop low duty cycle ultrasound pulsing strategies and nanocarriers to maximize non-thermal microvascular ablation and payload delivery to subcutaneous C6 gliomas.

Here, we describe a technique for the localized delivery of reagents to the rabbit mammary gland via an intraductal injection. In addition, we describe a protocol for visualization and the confirmation of delivery by high-resolution ultrasound imaging of contrast agents.
