Tri-layered Scaffold

A tri-layered scaffold is a bioengineered construct composed of three organized material layers, designed to reproduce the structural and functional complexity of native tissue. Each layer can provide distinct properties, such as differences in porosity, stiffness, composition, or degradation rate, while the interfaces between layers promote communication across the construct. This spatial organization helps regulate cell attachment, migration, proliferation, and differentiation within defined regions. In bioengineering, tri-layered scaffolds support the regeneration of tissues with multiple anatomical zones, including osteochondral and vascular structures, and provide a platform for studying how material architecture influences tissue formation and repair.

There has been renewed interest in developing polymer valves. Here, the objectives are to demonstrate the feasibility of modifying a commercial pulse duplicator to accommodate tri-leaflet geometries and to define a protocol to present polymer valve hydrodynamic data in comparison to native and prosthetic valve data collected under near-identical conditions.

For creation of highly organized structures of complex tissue, one must assemble multiple material and cell types into an integrated composite. This combinatorial design incorporates organ-specific layered cell sheets with two distinct biologically-derived materials containing a strong fibrous matrix base, and endothelial cells for enhancing new vessels formation.
