Circular Dna Template

A circular DNA template is a closed-loop DNA molecule used to preserve and copy genetic information for molecular biology experiments. DNA polymerases can repeatedly traverse the circle, and in rolling circle amplification, this continuous synthesis produces long, linked DNA copies from a small starting amount of template. In cancer research, circular DNA templates support the generation of defined genetic constructs, amplification of sequences containing cancer-associated variants, and development of sensitive assays for mutation or biomarker detection. Their stable, reusable format can improve experimental consistency and enable controlled studies of gene regulation, oncogenic mechanisms, and potential diagnostic methods.

Genome-wide Purification of Extrachromosomal Circular DNA from Eukaryotic Cells

This paper presents a sensitive method called Circle-Seq for purifying extrachromosomal circular DNA (eccDNA). The method encompasses column purification, removal of remaining linear chromosomal DNA, rolling-circle amplification and high-throughput sequencing. Circle-Seq is applicable to genome-scale screening of eukaryotic eccDNA and studying genome instability and copy-number variation.

This video demonstrates the circular dichroism (CD) spectroscopy technique to study conformational changes occurring in DNA in the presence of ATP-dependent chromatin remodeling protein. The changes in the CD spectra measured in the presence and absence of ATP hydrolysis indicate the importance of ATP in the ability of the remodeler to induce conformational changes in the bound DNA.
