Micropipette Alignment

Micropipette alignment is the precise positioning of a glass micropipette relative to a target cell or tissue, a critical step for obtaining reliable measurements in cancer research. In micropipette-based experiments, researchers align the pipette tip with the sample under microscopic control and apply controlled pressure or suction, minimizing off-axis contact and measurement error. Accurate alignment supports studies of cancer-cell deformability, membrane mechanics, adhesion, and interactions with surrounding tissues. These measurements can help characterize mechanical differences between healthy and malignant cells, improving understanding of tumor progression and informing research on cell invasion, metastasis, and treatment response.

Micropipette Aspiration of Substrate-attached Cells to Estimate Cell Stiffness

Here we describe a quick and simple method to measure cell stiffness. The general principle of this approach is to measure membrane deformation in response to well-defined negative pressure applied through a micropipette to the cell surface. This method provides a powerful tool to study biomechanical properties of substrate-attached cells.

We showcase the development of self-assembled, three-dimensional bundles of longitudinally aligned astrocytic somata and processes within a novel biomaterial encasement. These engineered "living scaffolds", exhibiting micron-scale diameter yet extending centimeters in length, may serve as test-beds to study neurodevelopmental mechanisms or facilitate neuroregeneration by directing neuronal migration and/or axonal pathfinding.
