Bacterial Staining

Bacterial staining is a group of laboratory methods that color bacterial cells to reveal their morphology, arrangement, and structural differences, supporting the study of infection and immune responses. In Gram staining, cells are treated with crystal violet and iodine, decolorized with alcohol, and counterstained with safranin; differences in peptidoglycan-rich cell walls determine whether bacteria appear purple or pink. Other stains can highlight capsules, spores, or acid-fast cell envelopes. These methods provide rapid visual evidence for identifying bacterial groups, guiding culture-based investigation, assessing clinical specimens, and linking microbial structure with pathogenicity and host defense.

DiRita1 1 Department of Microbiology and Molecular Genetics, Michigan State University, East Lansing, Michigan, United States of America Bacteria are diverse microorganisms found nearly everywhere on Earth. Many properties help distinguish them from each other, including but not limited to Gram-staining type, shape and arrangement, production of capsule, and formation of spores. To observe these properties, one...

Imaging InlC Secretion to Investigate Cellular Infection by the Bacterial Pathogen Listeria monocytogenes

Listeria monocytogenes is a Gram positive bacterial pathogen frequently used as a major model for the study of intracellular parasitism. Imaging late L. monocytogenes infection stages within the context of small-interfering RNA screens allows for the global study of cellular pathways required for bacterial infection of target host cells.
