Methanol Chloroform

Methanol-chloroform extraction is a solvent-based method for isolating lipids and other hydrophobic molecules from complex biological samples, making it useful in immunology and infection research. Methanol disrupts protein-lipid interactions and helps release cellular components, while chloroform dissolves nonpolar compounds; after water or aqueous buffer is added, the mixture separates into organic and aqueous phases that can be collected independently. Researchers use this approach to examine membrane lipids, microbial lipid components, and host lipid changes associated with infection or immune activation. The resulting extracts support biochemical analysis, lipid profiling, and studies of pathogen-host interactions.

We describe a new quantitative lipidomics method for identifying numerous lipid species in yeast using survey-scan electrospray ionization mass spectrometry (ESI/MS). This method exceeds currently available methods for lipid identification and quantification in the ability to resolve various molecular forms of lipids, sensitivity, and speed.
