Monocyte Dendritic Cell Analysis

Monocyte dendritic cell analysis is the characterization of monocytes, dendritic cells, and monocyte-derived dendritic cells to understand their identities, states, and roles in immune responses. Researchers typically combine cell isolation with flow cytometry, assessing surface markers such as CD14, CD16, HLA-DR, and CD11c, alongside morphology, cytokine production, and antigen-presenting capacity to distinguish related populations. In immunology and infection research, this analysis reveals how circulating monocytes differentiate, how dendritic cells capture and present microbial antigens, and how inflammatory signals shape their function. This video demonstrates the generation of immature, mature, and tolerogenic dendritic cells, or DCs, from human monocytes. The incubation of monocytes with growth factors and cytokines leads to the production of immature DCs. In response to immune regulatory molecules, immature DCs differentiate into tolerogenic and mature DCs.

This study compares two different methods of human monocyte isolation for obtaining in vitro dendritic cells (DCs). Monocytes are selected by adherence or negatively enriched by magnetic separation. Monocyte yield and viability along with MDDC viability, proliferation and CD11c/CD14 surface marker expression will be compared between both methods.

Dendritic cells (DCs) secrete IL-1β in response to TLR8 recognition of synthetic purine, R848, followed by NLRP3 inflammasome activation with nigericin, therefore, IL-1β can be used to measure NLRP3 inflammasome activity. Intracellular cytokine staining, immunoblotting, and ELISA are used to accurately measure NLRP3 inflammasome priming and activation via IL-1β expression.

This video demonstrates a protocol for assessing the activation of dendritic cells using immunoglobulin G (IgG)-coated tumor cells. Dendritic cells internalize the IgG-coated tumor cells, process the tumor antigens, and present them on the surface through MHC-II molecules, along with the co-expression of the costimulatory molecule CD86. These dendritic cells then undergo MHC-II and CD86-targeted immunostaining and flow cytometry analysis to identify their activation state.

We describe use of ImageStream technology (www.amnis.com), which combines quantitative flow cytometry with simultaneous high-resolution digital imaging, to quantify cellular mechanisms of primary immune cells from well-defined patient cohorts. Our studies provide a blueprint for translational investigations to quantify lineage specific cellular responses in small samples from subject cohorts.
