Double Dose Platelet

Double-dose platelet refers to a platelet component containing approximately two standard adult therapeutic doses, usually collected from one donor rather than pooled from multiple whole-blood donations. During plateletpheresis, an automated blood separator centrifuges anticoagulated blood, selectively retains platelets, and returns most red cells and plasma to the donor; collection settings may be adjusted according to donor size and platelet count. This approach can provide a higher, more consistent platelet yield for patients with thrombocytopenia or active bleeding. In medicine, it may reduce donor exposure, simplify inventory management, and support transfusion strategies when platelet demand is high, while requiring careful donor screening and quality control.

This article describes the process used by Örebro University Hospital to produce double dose buffy coat platelet concentrates prepared from whole blood donations and treated with the INTERCEPT Blood System for pathogen inactivation. The in vitro quality of the final platelet units are evaluated over 7 days of storage.

Here we show isolated human platelets can be used as an accessible ex vivo model to study metabolic adaptations in response to the complex I inhibitor rotenone. This approach employs isotopic tracing and relative quantification by liquid chromatography-mass spectrometry and can be applied to a variety of study designs.

We describe a straightforward method for the isolation of washed platelets from human blood followed by agonist-induced platelet aggregation measurements by turbidimetry. As an example we apply this method for studying the aggregation response of human platelets to collagen after a pre-incubation with the Pannexin1 channel inhibitor Brilliant Blue FCF.

This paper describes an experimental model of hemostasis that simultaneously measures platelet function and coagulation. Platelet and fibrin fluorescence is measured in real-time, and platelet adhesion rate, coagulation rate, and onset of coagulation are determined. The model is used to determine platelet procoagulant properties under flow in concentrates for transfusion.

Purification and Activation of Platelets from Murine Whole Blood

In this video, we describe a protocol for purifying platelets from murine whole blood and their subsequent activation with the platelet agonist thrombin. When analyzed with flow cytometry, the purified resting platelets are characterized based on the expression of their cell-surface marker CD41, whereas purified activated platelets co-express both the surface markers CD41 and CD62P.
