Perfusion Fluid Preparation

Perfusion fluid preparation is the controlled formulation of solutions used to circulate through blood vessels, organs, or tissues, where composition and sterility can directly affect experimental or clinical outcomes. It typically involves dissolving selected electrolytes, buffers, nutrients, or protective additives in a suitable solvent, then adjusting pH, osmolarity, temperature, and volume before filtration or other sterilization steps. In medicine, appropriately prepared fluids support tissue viability during organ preservation, extracorporeal circulation, and laboratory perfusion studies while helping limit cellular injury and unwanted coagulation. Careful preparation also improves reproducibility, enables comparison across perfusion systems, and supports safer translation of perfusion-based technologies.

Sample Preparation for Endopeptidomic Analysis in Human Cerebrospinal Fluid

A method for mass spectrometric analysis of endogenous peptides in human cerebrospinal fluid (CSF) is presented. By employing molecular weight cut-off filtration, chromatographic pre-fractionation, mass spectrometric analysis and a subsequent combination of peptide identification strategies, it was possible to expand the known CSF peptidome nearly ten-fold compared to previous studies.

Spatial Measurements of Perfusion, Interstitial Fluid Pressure and Liposomes Accumulation in Solid Tumors

The heterogeneous intra-tumoral accumulation of liposomes has been linked to an abnormal tumor microenvironment. Herein methods are presented to measure tumor microcirculation by perfusion imaging and elevated interstitial fluid pressure (IFP) using an image-guided robotic system. Measurements are compared to the intra-tumoral accumulation of liposomes, determined using volumetric micro-CT imaging.

Quantifying Single Microvessel Permeability in Isolated Blood-perfused Rat Lung Preparation

The isolated blood-perfused lung preparation makes it feasible to visualize microvessel networks on the lung surface. Here we describe an approach to quantify permeability of single microvessels in isolated lungs using real time fluorescence imaging.
