Skin Preparation Protocol

A Skin Preparation Protocol is a standardized sequence for cleansing and antiseptic treatment of the skin before a medical procedure, helping reduce contamination at the point of care. It typically involves inspecting the site, removing visible soil or oils, applying an appropriate antiseptic with controlled coverage, and allowing the area to dry for the required contact time; these steps lower the microbial burden without damaging the skin barrier. Used before injections, venipuncture, catheter placement, and surgery, the protocol promotes consistent technique, supports infection-prevention practice, and helps clinicians select preparation methods suited to the procedure and patient.

Frozen Skin Tissue Block Preparation: A Protocol to Preserve Cutaneous Melanoma Samples from Murine Dorsal Skin

2023

This video describes the detailed protocol for isolating melanoma-containing murine dorsal skin, followed by cryo-embedding to prepare a frozen skin tissue block. The cryo-embedded tissue block can be sectioned and microscopically examined to study melanoma progression.

CUBIC Protocol Visualizes Protein Expression at Single Cell Resolution in Whole Mount Skin Preparations

This report describes a CUBIC protocol to clarify full thickness mouse skin biopsies, and visualize protein expression patterns, proliferating cells, and sebocytes at the single cell resolution in 3D. This method enables accurate assessment of skin anatomy and pathology, and of abnormal epidermal phenotypes in genetically modified mouse lines.

An Improved Method for the Preparation of Type I Collagen From Skin

Traditional procedures for the isolation of soluble type 1 collagen (COL1) require about 10 days from start to finish because of lengthy buffer incubations and laborious resuspensions of fibrils. Here, we describe a means to purify COL1 from small dermal biopsies in less than 3 hr.

2017

This work presents a novel processing and imaging protocol for thick, three-dimensional tissue cross-section analysis that enables the full exploitation of confocal imaging modalities. This protocol preserves antigenicity and represents a robust system to analyze skin histology and potentially other tissue types.

The skin is home to a complex immune cell network. We describe an efficient methodology for the digestion of mouse skin, from different parts of the animal's body, in order to obtain a single-cell suspension and analyze the different leukocyte populations resident in the skin by flow cytometry.
