Tissue Slice Cultivation

Tissue slice cultivation is an ex vivo method that maintains thin sections of living tissue under controlled laboratory conditions, preserving key cellular interactions and aspects of native tissue organization. Slices are typically cut to allow diffusion of oxygen and nutrients from a culture medium, while carefully controlled temperature, sterility, and incubation conditions support viability and function. In medicine, this approach provides a practical model for studying tissue physiology, disease mechanisms, drug responses, and treatment toxicity. Because tissue slices retain multicellular architecture and local cell signaling, they can complement isolated cell cultures and help evaluate therapeutic strategies in a biologically relevant setting.

The video demonstrates the generation and maintenance of organotypic slice cultures from glioblastoma tissue. The isolated tissue is embedded in an agarose matrix, and thin sections are obtained using a vibratome. These sections are then placed on permeable membrane inserts within a microplate and cultured in a slice culture maintenance media for long-term maintenance.
