Cilium Marker Arl13b

Cilium marker Arl13b refers to the use of ADP-ribosylation factor-like protein 13B to identify primary cilia, slender, microtubule-based projections that organize signaling in many cells, including neurons. Arl13b is enriched in the ciliary membrane, where its localization can be detected by immunofluorescence and used to distinguish cilia from surrounding cellular structures. In neuroscience, Arl13b labeling helps researchers measure cilium number, length, and distribution in neural cells and brain tissue. These analyses support studies of neuronal development, cilium-dependent signaling, and disorders in which altered ciliary structure or function contributes to abnormal nervous-system development.

Using Primary Neurosphere Cultures to Study Primary Cilia

The primary cilium is fundamentally important in neural progenitor cell proliferation, neuronal differentiation, and adult neuronal function. Here, we describe a method to study ciliogenesis and the trafficking of signaling proteins to cilia in neural stem/progenitor cells and differentiated neurons using primary neurosphere cultures.

Recently we mapped the three-dimensional (3D) spatial locations of transport routes for various proteins translocating inside primary cilia in live cells. Here this paper details the experimental setup, the process of biological samples and the data analyses for the 3D super-resolution fluorescence imaging approach newly applied in live primary cilia.

This method describes the exploitation of the endogenous signal in an ARL13B-CENTRIN-2 transgenic mouse model to image primary cilia in situ, in three dimensions across large areas of mineralized tissue, from tissue collection to analysis of cilia organization. This protocol may also be applied to other types of tissues.

The Production of C. elegans Transgenes via Recombineering with the galK Selectable Marker

The ability to produce transgenes for Caenorhabditis elegans using genomic DNA carried by fosmids is particularly attractive as all of the native regulatory elements are retained. Described is a simple and robust procedure for the production of transgenes via recombineering with the galK selectable marker.
