Differential Protein Expression

Differential protein expression is the measurement and comparison of protein abundance between biological samples, conditions, or cell types to identify molecular changes. In neuroscience, researchers typically extract proteins from nervous-system tissues or cultured cells, quantify them using methods such as immunoblotting or mass spectrometry, and compare normalized signal intensities across experimental groups. Changes in protein levels can reveal altered signaling pathways, synaptic function, cellular stress responses, or disease-associated mechanisms. This approach supports the study of brain development, neurodegeneration, neuronal plasticity, and responses to drugs or injury, while helping connect molecular observations with cellular and behavioral outcomes.

Expression of malarial proteins in cell based systems remains challenging. We demonstrate two step and one step IVT (in vitro translation) cell free expression systems for expressing malarial recombinant rhoptry proteins from HeLa cells. We use a Ni-resin affinity based purification system to purify the rhoptry proteins.
