Human Brain Endothelial Cells

Human brain endothelial cells are specialized cells that line the blood vessels of the central nervous system and help regulate exchange between the bloodstream and neural tissue. Connected by tight junctions and supported by neighboring pericytes and astrocytes, they form the blood-brain barrier, which selectively controls transport of nutrients, signaling molecules, and potentially harmful substances. In neuroscience research, cultured human brain endothelial cells and barrier models help investigate vascular contributions to brain function, neuroinflammation, stroke, and neurological disease. These systems also support studies of drug permeability and delivery, providing a human-relevant platform for evaluating how therapeutics reach the brain.

An in vitro model for cerebral malaria sequestration is described1. Plasmodium falciparum infected red blood cells are selected for binding to immortalized human brain microvascular endothelial cells. The selected parasites show a distinct phenotype. The selection process can be applied using various P. falciparum strains and endothelial cell lines.

The aim of the protocol is to present an optimized procedure for the establishment of an in vitro blood-brain barrier (BBB) model based on primary porcine brain endothelial cells (pBECs). The model shows high reproducibility, high tightness, and is suitable for studies of transport and intracellular trafficking in drug discovery.
