Atomkraftmikroskopi (AFM) kombineret med scanning af elektrokemisk mikroskopi (SECM), nemlig AFM-SECM, kan bruges til samtidig at erhverve topografiske og elektrokemiske oplysninger i høj opløsning på materialeoverflader på nanoskala. Sådanne oplysninger er afgørende for at forstå heterogene egenskaber (f.eks. reaktivitet, defekter og reaktionssteder) på lokale overflader af nanomaterialer, elektroder og biomaterialer.
Scanning af elektrokemisk mikroskopi (SECM) bruges til at måle den lokale elektrokemiske adfærd af flydende / faste, flydende / gas og flydende / flydende grænseflader. Atomkraftmikroskopi (AFM) er et alsidigt værktøj til at karakterisere mikro- og nanostruktur med hensyn til topografi og mekaniske egenskaber. Konventionelle SECM eller AFM giver imidlertid begrænsede sideløste oplysninger om elektriske eller elektrokemiske egenskaber ved nanoskala. For eksempel er aktiviteten af en nanomaterialeoverflade på krystalfacetniveauer vanskelig at løse ved hjælp af konventionelle elektrokemimetoder. Dette papir rapporterer anvendelsen af en kombination af AFM og SECM, nemlig AFM-SECM, til sonde nanoskala overflade elektrokemiske aktivitet, mens erhverve høj opløsning topografiske data. Sådanne målinger er afgørende for at forstå forholdet mellem nanostruktur og reaktionsaktivitet, som er relevant for en lang række anvendelser inden for materialevidenskab, life science og kemiske processer. Alsidigheden af den kombinerede AFM-SECM demonstreres ved at kortlægge topografiske og elektrokemiske egenskaber af henholdsvis facetterede nanopartikler (NPs) og nanobubbles (NBs). Sammenlignet med tidligere rapporterede SECM-billeddannelser af nanostrukturer muliggør denne AFM-SECM kvantitativ vurdering af lokal overfladeaktivitet eller reaktivitet med højere opløsning af overfladekortlægning.
Karakterisering af elektrokemisk (EF) adfærd kan give kritisk indsigt i kinetik og mekanismer af interfaciale reaktioner inden for forskellige områder, såsom biologi1,2, energi3,4, materialesyntese5,6,7og kemisk proces8,9. Traditionelle EF-målinger, herunder elektrokemisk impedansspektroskopi10, elektrokemiske støjmetoder11, galvanostatisk intermitterende titrering12og cyklisk voltammetri13 udføres normalt i makroskopisk skala og giver en overfladegennemsnitsrespons. Det er således vanskeligt at udtrække oplysninger om, hvordan elektrokemisk aktivitet fordeles over en overflade, men lokale overfladeegenskaber i nanoskala er særligt vigtige, hvor nanomaterialer anvendes i vid udstrækning. Derfor er nye teknikker, der samtidig kan opfange både nanoskala multidimensional information og elektrokemi, meget ønskelige.
Scanning af elektrokemisk mikroskopi (SECM) er en udbredt teknik til måling af materialers lokaliserede elektrokemiske aktivitet ved mikro- og nanoskalaer14. SECM bruger typisk et ultramikrobiktrode som en sonde til påvisning af elektroaktive kemiske arter, når det scanner en prøveoverflade for at løse lokale elektrokemiske egenskaber15. Den målte strøm ved sonden frembringes ved reduktion (eller oxidation) af mæglerarten, og denne strøm er en indikator for den elektrokemiske reaktivitet på prøvens overflade. SECM har udviklet sig betydeligt efter sin første start i 198916,17, men det er stadig udfordret af to hovedbegrænsninger. Da EF-signaler typisk er følsomme over for karakteristika for interaktion mellem spids og substrat, er en begrænsning af SECM, at det at holde sonden i konstant højde forhindrer en direkte korrelation mellem elektrokemisk aktivitet og overfladelandskabet på grund af sammenkædningen af topografi med de indsamlede EF-oplysninger18. For det andet er det vanskeligt for et kommercielt SECM-system at opnå submikrometer (μm) billedopløsning, da den rumlige opløsning delvis bestemmes af sondedimensionerne, som er på mikrometerskalaen19. Derfor anvendes nanoelektroder, elektroderne med en diameter i nanometerområdet i stigende grad i SECM for at opnå en opløsning under submikrometerskalaen20,21,22,23.
For at give en konstant tip-substrat afstandskontrol og opnå en højere rumlig elektrokemisk opløsning er der anvendt flere hybride teknikker til SECM, såsom ionledningsføringspositionering24, forskydningskraftpositionering25, vekselstrøm SECM26og atomkraftmikroskopi (AFM) positionering. Blandt disse instrumenter er SECM’s integration af AFM-positionering (AFM-SECM) blevet en meget lovende tilgang. Da AFM kan levere faste spids-substrat afstande, den integrerede AFM-SECM teknik muliggør samtidig erhvervelse af nanoskala overflade strukturelle og elektrokemiske oplysninger gennem kortlægning eller prøve fejer med de skarpe AFM tips. Siden MacPherson og Unwins første vellykkede drift af AFM-SECM i 199627er der opnået betydelige forbedringer med hensyn til sondedesign og -fremstilling samt dets anvendelser inden for forskellige forskningsområder såsom elektrokemi i kemiske og biologiske processer. For eksempel er AFM-SECM blevet implementeret til billeddannelse af kompositmaterialeoverflader, såsom ædle metalnanopartikler28, funktionaliserede eller dimensionelt stabile elektroder29,30og elektroniske enheder31. AFM-SECM kan kortlægge de elektrokemisk aktive steder fra spidsen aktuelle billede.
Samtidige topografiske og elektrokemiske målinger kan også opnås ved hjælp af andre teknikker såsom ledende AFM32,33,34,35, elektrokemisk AFM (EC-AFM)36,37,38,39, scanning ionledningsføringsmikroskopi-scanning af elektrokemisk mikroskopi (SICM-SECM)24,40og scanning af elektrokemisk cellemikroskopi (SECCM)41,42 Sammenligningen mellem disse teknikker er blevet diskuteret i et gennemgangsdokument1. Formålet med dette arbejde var at anvende SECM-AFM til at demonstrere den elektrokemiske kortlægning og måling af facetterede krystallinske akvariske oxid nanomaterialer og nanobobler i vand. Facetterede nanomaterialer syntetiseres bredt for metaloxidkatalysatorer i rene energiapplikationer, fordi facetterne med karakteristiske krystallografiske træk har karakteristiske overfladeatomstrukturer og yderligere dominerer deres katalytiske egenskaber. Desuden målte og sammenlignede vi også den elektrokemiske adfærd ved væske-/gasgrænsefladerne for overflade nanobubbles (NBs) på guldsubstrater. NBs er bobler med en diameter på <1 μm (også kendt som ultrafine bobler)43, og de fremkalder mange spændende egenskaber44,45, herunder lange opholdstider i løsningerne46,47 og høj effektivitet af gasmasseoverførsel46,48. Desuden skaber sammenbruddet af NBs chokbølger og dannelsen af hydroxylradikaler (•OH)49,50,51,52. Vi målte den elektrokemiske reaktivitet af ilt-NBs i opløsningen for bedre at forstå NBs grundlæggende kemiske egenskaber.
En kombineret AFM-SECM-teknik, der muliggør multimodal billedbehandling med høj opløsning, er beskrevet i denne protokol. Denne teknik gør det muligt at kortlægge topografi samtidig med SECM-strømmen, der er indsamlet eller kortlagt på enkelte nanopartikler eller nanobubbles. Eksperimenter blev udført ved hjælp af kommercielle sonder.
