Nematode Concentration Adjustment

Nematode concentration adjustment is the process of changing the number of nematodes suspended in a sample to achieve a defined density for biological experiments. It typically involves mixing the suspension thoroughly, measuring nematode abundance in a representative aliquot, and adding a suitable medium or removing excess liquid until the target concentration is reached. Accurate adjustment improves consistency across treatments, supports reliable dosing in infection, toxicity, behavioral, and developmental assays, and helps researchers compare outcomes between experiments. Careful handling also limits settling and uneven distribution, which can otherwise introduce variation and reduce the reproducibility of nematode-based measurements.

Visualizing Bacteria in Nematodes using Fluorescent Microscopy

To study the mutualism between Xenorhabdus bacteria and Steinernema nematodes, methods were developed to monitor bacterial presence and location within nematodes. The experimental approach, which can be applied to other systems, entails engineering bacteria to express the green fluorescent protein and visualizing, using fluorescence microscopy bacteria within the transparent nematode.

Behavioral assays provide powerful tools for understanding neuronal function. Here we present several protocols for quantifying predatory feeding behavior found in the model nematode Pristionchus pacificus and its relatives. Additionally, we provide methods for analyzing predatory feeding adaptations including mouth structures and teeth.

An In Vivo Assay to Detect Mitophagy in Transgenic Nematodes

This video demonstrates an in vivo assay to study mitophagy in Caenorhabditis elegans. Upon exposing the nematodes to a mitophagy-inducing drug, the occurrence of mitophagy is assessed by co-localization between DCT-1, a mitophagy receptor on the damaged mitochondria, and the autophagosomal membrane protein LGG-1.
