Chemokine Injection

Chemokine injection is an experimental method that delivers chemokine proteins into a tissue or organism to examine how these signaling molecules control immune-cell movement. After administration, chemokines bind to matching G protein-coupled receptors on responsive cells and establish local concentration gradients that guide chemotaxis toward the injection site. In biology research, this approach helps characterize leukocyte recruitment, inflammatory signaling, and tissue-specific immune responses. By measuring cell migration and changes in the surrounding tissue, researchers can assess chemokine function and investigate processes relevant to infection, wound repair, inflammation, and immune regulation.

The three-dimensional flow chamber device is a novel in vitro technology for the quantitative and step-wise evaluation of the extravasation cascade of cells circulating under conditions of physiological shear stress. To test the effect of a chemokine on macrophage recruitment in vivo, the whole mount in situ hybridization was used to detect the ectopic expression of the chemokine, and immunostaining was used to label macrophages. Live imaging was used for real-time observation of macrophage migration.
