Mouse Hippocampal Neurons

Mouse hippocampal neurons are nerve cells isolated from the hippocampus of mice, a brain region involved in learning, memory, and spatial information processing. They receive and transmit electrical and chemical signals through dendrites, axons, and synapses, while activity-dependent changes in synaptic strength support neuronal communication and plasticity. In laboratory studies, cultured mouse hippocampal neurons provide an accessible model for examining neuronal development, morphology, electrophysiological activity, and synaptic function. Researchers use them with imaging, molecular assays, and drug treatments to investigate mechanisms of neural signaling, neurotoxicity, and neurological disease, helping connect cellular processes with broader biological and behavioral outcomes.

2025

The video demonstrates a method for culturing neurons from murine hippocampi. Initially, the hippocampi undergo enzymatic digestion to break down the extracellular tissue matrix. Subsequently, the digested tissue is mechanically disrupted to isolate individual neurons. Finally, these neurons are cultured on glial feeder cells.

This protocol outlines the steps required to dissect, transfect via electroporation and culture mouse hippocampal and cortical neurons. Short-term cultures may be used for studies of axon outgrowth and guidance, while long-term cultures can be used for studies of synaptogenesis and dendritic spine analysis.
