Perforin Granzymes

Perforin granzymes are a coordinated set of effector proteins used by cytotoxic T lymphocytes and natural killer cells to eliminate infected, damaged, or abnormal target cells. After immune recognition and formation of an immunological synapse, perforin disrupts the target-cell membrane, allowing granzymes, which are serine proteases, to enter and activate apoptosis through intracellular substrates and signaling pathways. This mechanism supports immune surveillance and defense against viral infections while limiting collateral damage through targeted cell killing. Abnormal perforin-granzyme activity is associated with immunodeficiency, autoimmune disease, and cancer, making the pathway important in studies of immune regulation and immunotherapy.

We describe a simple, quantitative colorimetric assay that specifically measures the proteolytic activity of human, mouse or rat Granzyme B (GzmB). This protocol can be easily adapted for determining protease activity of other granule serine proteases by the hydrolysis of other synthetic peptide substrates with an appropriate recognition sequence.

This video demonstrates an ex vivo technique to induce brain cell death mediated through cytotoxic CD8+ T cells. Upon obtaining brain slices, activated oligodendrocyte-directed CD8+ T cells are transferred onto the slices to target the myelin sheath and oligodendrocytes. Post-incubation, the damage to myelin and neuronal death can be assessed histologically in the regions of interest.
