Peptide Isomer Separation

Peptide isomer separation is the analytical process of distinguishing peptides that share the same composition but differ in sequence, stereochemistry, or three-dimensional structure. It works by exploiting small differences in properties such as charge, hydrophobicity, conformation, and interactions with a stationary phase, often through liquid chromatography and, when needed, ion-mobility or mass spectrometric analysis. In chemistry, separating these closely related species supports peptide identification, purity assessment, and structural characterization. The approach is important in proteomics, pharmaceutical quality control, and studies of how isomeric peptides can produce different biological effects.

Free solution capillary electrophoresis is a fast, cheap and robust analytical method that enables the quantitative monitoring of chemical reactions in real time. Its utility for rapid, convenient and precise analysis is demonstrated here through analysis of covalent peptide grafting onto chitosan films for improved cell adhesion.

We present a technique that allows the spatial separation of different conformers or clusters present in a molecular beam. An electrostatic deflector is used to separate species by their mass-to-dipole moment ratio, leading to the production of gas-phase ensembles of a single conformer or cluster stoichiometry.
