Pdgfr Alpha Positive Cells

PDGFR alpha positive cells are cells that express platelet-derived growth factor receptor alpha, a cell-surface receptor that helps regulate proliferation, migration, survival, and differentiation during tissue development. When platelet-derived growth factor binds the receptor, PDGFR alpha activates intracellular signaling pathways that influence cell behavior and responses to the surrounding tissue environment. In developmental biology, these cells contribute to the formation and maturation of mesenchymal tissues, including connective tissue, cartilage, and other organ structures, depending on their location and developmental stage. Identifying PDGFR alpha positive populations helps researchers trace cell lineages, characterize progenitor cells, and investigate how altered signaling affects development and disease.

Source: Tagliafierro, L., et al. Lentiviral Vector Platform for the Efficient Delivery of Epigenome-editing Tools into Human Induced Pluripotent Stem Cell-derived Disease Models. J. Vis. Exp. (2019)This video demonstrates the lentiviral transduction of neural progenitor cells (NPCs) for targeted epigenetic modification of alpha-synuclein, a key protein in Parkinson’s disease. The protocol enables methyltransferase-mediated DNA methylation, reducing SNCA expression for studying gene regulation...

Induction of Neuroinflammation in Mice via Injection of Alpha-Synuclein Fibrils

2025

This video demonstrates the injection of alpha-synuclein fibrils into a mouse to induce neuroinflammation. The fibrils are injected intraperitoneally into a transgenic mouse expressing mutated alpha-synuclein proteins. Upon reaching the central nervous system, the fibrils enter neurons and induce aggregation of the mutated proteins. The aggregates spread to neighboring neurons, microglia, and astrocytes. Recognition of the aggregates triggers microglia to induce neuroinflammation, damaging the...

Immunostaining of Neurons Treated with Alpha-Synuclein Aggregates

2025

This video demonstrates the immunostaining of neurons treated with alpha-synuclein (αS) aggregates. The neurons are first treated with αS aggregates. They are then fixed, permeabilized, and treated with a blocking solution. Primary antibodies are added, followed by a fluorescently labeled secondary antibodies and a nuclear stain. The sample is then mounted and observed under a microscope.
